چکیده

55th ASMS Conference – Indianapolis 2007 www.chemistry.wustl.edu/~msf/ Overview Purposes •Evaluate the resolving power and sensitivity achievable with a new compensated trap for FTMS as a function of m/z and develop a protocol for tuning this trap. Methods •Sample empirically the cyclotron frequency surface at various cyclotron radii and z-mode amplitude distributions. •Compare the mass resolving power and sensitivity at various m/z’s achievable with a compensated trap compared to those achievable with the conventional (uncompensated) trap. Results •Compensated trap achieves nearly three fold increase in apparent resolving power and a ten fold increase in sensitivity. •An experiment-based algorithm flattens the frequency surface of the new trap in one step. •The flattened surface, achieved by applying voltages to auxiliary rings, insures a more constant ion frequency as a function of ion mode amplitudes, than does uncompensated trap.

The two human centrin homologues have similar but distinct functions at Tetrahymena basal bodies

Centrins are a ubiquitous family of small Ca(2+)-binding proteins found at basal bodies that are placed into two groups based on sequence similarity to the human centrins 2 and 3. Analyses of basal body composition in different species suggest that they contain a centrin isoform from each group. We used the ciliate protist Tetrahymena thermophila to gain a better understanding of the functions ...

Differential expression and cellular distribution of centrin isoforms during human ciliated cell differentiation in vitro.

Centrin protein is an ubiquitously expressed cytoskeletal component and is a member of the EF-hand superfamily of calcium-binding proteins. It was first discovered in the flagellar apparatus of unicellular green algae where it is involved in contraction of Ca(2+)-sensitive structures. Centrin protein is associated with centrosome-related structures such as spindle pole body in yeast, and centri...

Expression of centrin isoforms in the mammalian retina.

Centrin is a calcium-binding phosphoprotein of centrosomes, mitotic spindle poles, and flagellar basal apparatus. Indirect immunofluorescence studies in human and rat retinas reveal centrin localization in two distinct cellular structures: at centrosomes of nonciliated neuronal cells as well as in basal bodies, and in larger amounts in the highly modified cilium--the connecting cilium--of photo...

CK2 phosphorylation of human centrins 1 and 2 regulates their binding to the DNA repair protein XPC, the centrosomal protein Sfi1 and the phototransduction protein transducin β

Centrins are calcium-binding proteins that can interact with several cellular targets (Sfi1, XPC, Sac3 and transducin β) through the same hydrophobic triad. However, two different orientations of the centrin-binding motif have been observed: W(1)xxL(4)xxxL(8) for XPC (xeroderma pigmentosum group C protein) and the opposite orientation L(8)xxxL(4)xxW(1) for Sfi1 (suppressor of fermentation-induc...

Cloning, localization, and axonemal function of Tetrahymena centrin.

Centrin, an EF hand Ca(2+) binding protein, has been cloned in Tetrahymena thermophila. It is a 167 amino acid protein of 19.4 kDa with a unique N-terminal region, coded by a single gene containing an 85-base pair intron. It has > 80% homology to other centrins and high homology to Tetrahymena EF hand proteins calmodulin, TCBP23, and TCBP25. Specific cellular localizations of the closely relate...

Structural role of Sfi1p–centrin filaments in budding yeast spindle pole body duplication

Centrins are calmodulin-like proteins present in centrosomes and yeast spindle pole bodies (SPBs) and have essential functions in their duplication. The Saccharomyces cerevisiae centrin, Cdc31p, binds Sfi1p on multiple conserved repeats; both proteins localize to the SPB half-bridge, where the new SPB is assembled. The crystal structures of Sfi1p-centrin complexes containing several repeats sho...
